OBJECTIVE To study the effect of high-fat diet on the expression of MMP-9 and PPAR α in rat's aorta.METHODS Male Wistar rats were randomly divided into control group , high-fat diet group (HF) and Fenofibrate -treated group (HF + FF). Fenofibrate was administrated orally for 20 weeks, plasma triglyceride and cholesterol levels were measured. Protein levels and mRNA levels of MMP-9 and PPAR α of rats' aorta were determined using Western blot and RT-PCR.RESULTS Compared with control, plasma triglyceride and cholesterol levels increased significantly in HF and HF+FF (T= 3. 69-9. 32 , P<0.05),and the level of TC HDL-C LDL-C is higher, in HF than in HF+FF(T= 2. 93~5. 24 , P<0. 05). High-fat diet increased both the mRNA and protein levels of MMP-9 in rats aorta (T= 4. 65~5. 48 , P < 0.01), accompanied with the decreased expression of PPARa (T= 2. 52~2. 98 , p<0.05). Compared with HF group, administration of Fenofibrate could increase both the mRNA (t= 4. 51—6. 29 , P<0. 01), and protein expression (t= 5. 62~5. 97, P<0. 01) of PPAR α , accompanied with amelioration of high fat diet-induced increase of MMP-9.CONCLUSION 1.High fat diet can lead to higher plasma triglyceride and cholesterol levels directly. 2.Higher plasma triglyceride and cholesterol levels impairs both the mRNA and protein expression of PPAR α and increase the expression of MMP-9.3. Administration of Fenofibrate could ameliorate the disturbance of MMP-9 possibly via a PPARa mediated mechanism.4.There is a certainly relationshiop between expression of PPAR α and MMP-9.5.Through the way above. Fenofibrate provide possibly protection to aorta by the effort to arteriosclerosis.
