Objective:1. Ovarian carcinoma was the leading cause of death for malignancies of the female genital tract. Two kinds of gene could be changed during the development of cancer at least, that were activation of oncogene and mutational activation or deletion of tumour suppressor gene. FHIT gene was a new be identified as tumour suppressor gene recently. In this study, the nested PCR could gain the transcript of the exons 3-10 of gene FHIT to research the expressions of FHIT gene with the development of ovarian carcinoma.2. Loss of heterozygosity (LOH) was the main reason cause the mutational activation of tumour suppressor gene. Single strand conformation polymorphism analysis of polymerase chain reaction products (PCR-SSCP) is a kind of method for analysis of DNA recently. In this study, we were to detect the possible reason of inactivation of suppressor genes though PCR-SSCP which were based primarily on LOH near introns 5 of FHIT gene, thus to elucidate the role of FHIT in the development of ovarian carcinoma.Method:1. Two pairs of PCR primers were used for Nested PCR in this study, to research the expressions of FHIT gene in ovarian epithelial carcinoma tissues and normal ovarian tissues.
