Objective To observe the effect of hypoxia-inducible factor 1 a following the global cerebral ischemia in rats, And to observe the treatment effect of 2ME2 on hypoxia-inducible factor 1 a and the gene (RTP801, caspase-3) involved in apoptosis.Methods Ninety adult male Sprague - Dawley rats were randomly divided into 3 groups: sham group (n=6), global cerebral ischemia group(GI, n=42) and global cerebral ischemia + 2ME2 treament group (GI+2ME2, n=42), and the rats of GI group and GI+2ME2 group were sacrificed at 6 hour, 12 hour, 24 hour, 48 hour, 96 hour, 5 day and 7 day after perfusion. The 4 vessel occlusion (4-VO) global ischemia was produced. The electroencephalogram (EEG) was recorded by powerlab system. Nissl staining was used to perform quantitative analysis of cell numbers. Immunohistochemistry staining and RT-PCR will provide information of the distribution of positive cells to HIF-1a proteins caspase-3 protein and the expression of RTP801mRNA respectively. The rats in GI+2ME2 group were treated with 2ME2 by abdominal cavity injection (16mg/ kg /day) and one time a day with the same dosage in the first 3 days.
