Objectives To investigate the effects of Valproic acid(VPA) on the proliferation of human ovarian carcinoma cell line A2780 and the expression of angiogenesis and metastasis related factors(VEGF , MMP-9 , MMP-2 and E-cadherin).Methods Human ovarian carcinoma cell line A2780 cells were cultured in vitro and exposed to VPA in different concentration(0,1,2, 3,4, 5 mmol/L) for 24~72h. The proliferation, morphological changes and the characters of apoptosis of the cells were observed by inverted microscope and HE staining. The proliferative activities(OD mean value and growth inhibite rate) of cells were measured by MTT. The characters of apoptosis and cycle of the cells were assessed by flow cytometric analysis. Expression of VEGF, MMP-9, MMP-2 and E-cadherin was assayed by immunocytochemistry and flow cytometric analysis.Results The proliferation and morphological changes were obviously different compared with that of the control group. Cells of the control group grew well and proliferated continually, while cells treated with VPA grew slowly and had obvious morphological changes. The inhibition of proliferation of A2780 cells by VPA was in a dose-time depended fashion. The mount of cells...
