OBJECTIVE: To investigate the effects of valproic acid(VPA) on proliferation and expression of vascular endothelial growth factor(VEGF) , E-cadherin and matrix metalloproteinase-2, 9 (MMP-2, MMP-9 ) in ovarian cancer cell line SKOV3 cells.METHODS: Cultured SKOV3 cells was treated by VPA with different concentration and time.The morphological changes of SKOV3 cells treated by VPA was observed through inverted microscope and the proliferation was predicted by methyl thiazolyl tetrazolium (MTT). The characters of apoptosis were assessed by flow cytometric analysis.The expression of VEGF, E-cadherin, MMP-2 and MMP-9 protein was observed by immunocytochemistry(IC). The VEGF , E-cadherin, MMP-2 and MMP-9 protein level was determined by Flowcytometry.RESULTS: (1) Under light microscope, cells in the controle group grew bloomly and were clustered. The shapes were long ,shuttle and polygon. The cell shapes were plump.Cells were abundant in cytoplast.The cell nucleus were in the middle of the cell.When the concentration of VPA was less than 2mmol/L,the growth of cells in the experimental group were not inhibited evidently.As the concentration of VPA rise to 3mmol/L or more,the growth of the cells was inhibited and the normal cells were dropped off.The residual attached cells were scattered or rickled.The shape of the residual attached cells were round or ellipse.The cytoplast were crimpled.(2) When the concentration of VPA was between 3 to 5 mmol/L,the growth of SKOV3 cells was inhibited evidently in vitro.After using VPA for 72h, the cell...
