[Objective] : By setting up rat models of alcoholic fatty liver, To observe the pathology and ultramicro-structure changes, and to investigate the proteasome LMP7 mRNA expression and protein content, also to investigate the variation of liver damage index and anti-oxidation index, and to treat with tiopronin,Thus to reveal the mechanism of alcoholic liver disease and to provide experiment basis for clinical therapy.[Methods] : One hundred male Wistar rats weighting between 170g and 190g were fed for one week. Then divided into two groups randomizedly: model group and control group, 80 rats in the model group and 20 in the other. The model group was given alcoholic intragastric administration plus Oliver oil diet. The diet supplied 18% of the total calories as protein, 14% as fat, 12% as carbohydrate and 46% as ethanol; also vitamin compounds and essential mineral matter were given. The ethanol was infused chronically, in the first 4 weeks, 30% ethanol was infused by 4g · Kg-1 · d-1,in the next 4 weeks, 40% ethanol was infused by 5g · Kg-1 · d-1, then in the last 4 weeks, 50% ethanol was infused by 6g · Kg-1 · d-1, with ethanol inner-gastric infused twice a day. The rats of model group were divided into three sub-groups randomizedly: one group was given 60% ethanol 9g · Kg-1 · d-1 plus tiopronin 300mg · Kg-1 · d-1, one group was given 60% ethanol 9g·Kg-1·d-1, another group was given equal normal sodium. That is treatment group, untreated group and normal sodium group. All the groups were treated for four weeks. Then they were killed by decapitation.
