It has been proved by former researches that acute leukemia is a tumour relating to multi-genes. Cloning a novel leukemia associated gene is one hotspot in the field. it not only facitiliate to explore the mechanism of malignant hematopoietic diseases,but also to find new target sites for therapy.【Objective】To clone the full-length cDNA of a novel gene of familial acute myelogenous leukemia and to explain the molecular mechanisms of the disease at the gene level.【Method】our laboratory constructed a subtractive cDNA pool of familial acute myelogenous leukemia using the method of suppression subtractive hybridization (SSH) in 2002 and obtained 28 clones; 28 clones were sequenced and compared with known sequences in the public databases of GenBank/EMBL/DDBJ using NCBI BLAST for homology analysis,Then found that 17 sequences were a part of known genes, and the rests were verified unknown fragments;detecting mRNA of the rest 11 unknown sequences in familial acute myelogenous leukemia patient sample and health adult sample with one-step semi-quantitative reverse transcriptase-PCR(RT-PCR) in order to exclude the possibility of false positivity from SSH, 4 sequences were confirmed to be positive differentially expressed genes fragments. On the basis of the 4 sequences ,the experiment was designed as follow: 1. clone the full-length cDNA of a novel associated gene of familial acute myelogenous leukemia by the electronic cDNA cloning and SMART-rapid amplification of cDNA ends(SMART-RACE); 2. predict their associated function by bioinformatics strategy.【Results】One sequence of 2257 bp was obtained with complete ORF and polyA tail ,and localized on 4q31.1 of human chromosome; the ORF ranged between150~581nt of the gene and coded a...
