Objective To study the mechanism of antler polypeptides promoting chondrocytes proliferation in cellular and molecular lever, we investigate the critical molecules on the signal transductional way such as receptor tyrosine kinase. Proto-oncogene c-fos, CyclinA. Methods Cultured rabbit chondrocytes were stimulated by various concentrations of antler polypeptides and genistein, a tyrosine kinase inhibitor, Cell proliferation were determined by MTT assay and Flow cytometry. SABC techniques were applied for CyclinA immunocytochemical staining. The expression of c-fos mRNA was assessed by RT-PCR. Results Cell proliferation was promoted by antler polypeptides with increasing proliferation percentage and more cells in S stage of cell cycle, up-regulating the percentage of positive staining CyclinA cells and expression of c-fos m RNA. But Genistein suppressed the proliferation with decreasing percentage in S stage, Both of the two substances showed dose-dependent manners. Conclusion Antler polypeptides stimulate the proliferation of chondrocytes by inducing more S-stage entry via tyrosine kinase pathway, which induced the expression of proto-oncogenes c-fos responsible for modulating CyclinA...
