Objective:To Screen for active ingredient against hepatic cell injured by CCl4 in Rubus alceafolius Poir through activity-guided tracing methods.To set up preliminary identification of active ingredient and study the possible mechanism of the action.Methods:(1)The general alkaloid of Rubus alceaefolius Poir were extracted by traditional Chinese medicine routine and detected.(2) According to differences of polarity in general alkaloid,menstruum such as respectively,using ethyl acetate,n-butanol,the chloroform fight extract whose different organic solvents fractions.(3)Establishing a hepar-injured model and interventioning with different organic solvents fractions in Rubus alceaefolius Poir,and observing the effects of the different organic solvents fractions,and choosing the active organic solvents fractions against Hepatic injury.(4)By the methods of traditional column chromatography,the active organic solvents fractions was divided further,and merging the same or similar fraction of distillate.(5)Dividing the componts by large-scale chromatography and HPLC further.(6)Culturing the hepatic cell of rat,and establishing model of hepatic cell damaged.Interventioning the model with active ingredient and observeing the change of correlated indexes.The active ingredient in Rubus alceaefolius Poir against hepatic injury was evaluated. called 129③,whose different concentration could degrade the contents of ALT,AST in supernate fluid of cultured cell differently;(2) 129③,whose different concentration could heighten the activity of SOD in supernate fluid of cultured cell differently,and degrade the content of MDA.(3) 129③,whose different concentration could promote the proliferation of hepatic cell differently.(4) 129③,whose different concentration could regulate the cell cycle,and precipitate hepatic cell to S period.(5) 129③,which could regulate down the mRNA expression of Caspase-3 and regulate up the mRNA expression of PCNA significantly.Conclusion:(1) 129③is the active ingredient in Rubus alceaefolius Poir against hepatic injury.(2) 129③could protect the cellular membrane,lessen the degree of hepatocyte injured.(3) 129③could restrain the free radical coming into being and lipid peroxidation outcome coming into being,and improve liver cells antioxidant capacity.(4) 129③could boost the hepatocyte proliferating,restrain apoptosis of a cell.(5) 129③could regulate the cell cycle.
