| Objective: To investigate the differentiation of rat's bone marrow stromal cells(BMSC)into nerve cells with Acanthopanax Senticosus Injection. Methods: The bone marrow stromal cells of Wistar rat were separated and purified by percoll. The BMSCs was induced by 100μl/ml Acanthopanax Senticosus Injection . The differentiation was observed under inverted microscope; the expression of neuronal and glia specific markers such as NSE,MAP-2 and GFAP and genes such as NCAM,GAP43,SYN1 and GFAP was evaluated by immunocytochemistry cytoche mistry staining and reverse transcriptase- polymerase chain reaction(RT-PCR). Results: After 6 hours of induction, cytoplasm of BMSC contracted with protruding process, prominences form network structure after 24 hours. The immunocytochemical staining was performed on cells after induction , the differentiating cell expressed NSE and MAP-2, but negative in GFAP immunocyto- chemical staining; expression of neuron-specific makers and genes such as NCAM,SYN1 and GAP43 was detectable, and glial-specific markers and gene such as GFAP was not expressed. Conclusion: Acanthopanax Senticosus Injection could induce rat BMSCs differentiate into nerve cells in vitro. |
