Experimental Studies Of Acanthopanax Senticosus Injection Induced Bone Marrow Stromal Cells Transplanted For Focal Cerebral Ischemia Therapy

Objective:This study was to observe the treatment and mechanisms of acanthopanax senticosus injection induced bone marrow stromal cells(BMSC) transplanted for focal cerebral ischemia in rat. Methods:Wistar rats were randomly divided into four groups: the sham-operated groupthe pure cerebral ischemia group,bone marrow stromal cells group, bone marrow stromal induced by acanthopanax senticosus injection group. Rats in sham-operated group were only exposed and separated blood vessel and nerve.The pure cerebral ischemia group was established by right middle cerebral artery occlusion(MCAO). And the other groups:the BMSCs group,in which the rats were transplanted BMSCs,and the BMSCs induced by acanthopanax senticosus injection group, in which the rats were transplanted induced BMSCs. The nerves function score was determined by Chen,s Neurological Severity Scores method. The brain,s HE and Nissl staining pathology construction was observed by light microscope. The expression of BDNF and survival of transplanted cells were detected by the immunohistochemical method. SPSS13.0 statistical software was applicated to detect statistical number , and complete randomly single-factor analysis of variance (ANOVA)was done, comparing the differences between different groups .The difference between the groups had statistical significance(P≤0.05).Results:The nerves function scores of each group in which BMSCs were tansplanted were less than the pure cerebral inchemia group .Significant recovery of function was found in rats treated with induced BMSCs compared with BMSCs group. Conclusion:The transplantation of BMSCs can lessen the neurologic impairment symptom caused by cerebral ischemia damge.
