Objective: to study the correlation of polymorphism of the DNA repair gene- XRCC1 and susceptibility to gastric cancer in Shandong province. Methods: A 1:1 case-conrol study of 61 patients in Shandong province with primary gastric cancer and free-cancer or alimentary inpatients as a control group (matched for ages±5 years and sex) was conducted to detect XRCC1 polymorphism at both G27466A and G28152A . genetypes were analysed by PCR-based restriction fragment length polymorphism techniques.all of them were examined in epidemiology ,including history of smoking,drinking and family history of tumor.We drawn 2ml peripheral blood from subjects,separated DNA,compared different genetypes with the relationship of gastric cancer and analysed the interaction of XRCC1 polymorphism and environmental factors in the development of gastric cancer. RESULTS:The frequency of XRCC1-280His allele gene in cases and controls were 8.20% and 16.32% separately,the different was insignificant. The frequency of XRCC1-399Gln allele gene in cases and controls were52.46% and 40.98% separately,the different was insignificant.However,the subjects with 399Gln allele gene have a higher risk in gastric cancer(OR=2.708,95%CI=1.109~6.615,P=0.027).There is no synergy in the two points of XRCC1--G27466A and G28152A,Which have no mutualization with smoking and drinking. Conclusions: G28152A polymorphism of XRCC1 gene may increase susceptibility to gastric cancer in male,and have no mutualization with smoking and drinking.
