Eupolyphaga sinensis Walker is a kind of insects that widely used to be a drug in the fields of Chinese traditional medicine. With the modern biochemistry theory and isolating technology, we used fresh female Eupolyphaga sinensis Walkers to be raw material, decocted with distilled water under 90℃, sedimentated with ethanol under 4℃, removed the protein with Sevag reagent. Two groups of Glycoprotein were separated after DEAE-50 chromatogram, group one was brine washed with 0.4mol/L NaCl, another was with lmol/L. We analyzed the Glycoprotein group one with PAGE and SDS-PAGE, the results suggest that the Glycoprotein group one contains at less four different components. Consulted to the protein mark, the molecular weight of each component is 36.4KD, 42.8KD, 48.4KD and 53.3KD.We used two human cancer cell species: Eca109 and Hela to analyze the anti-cancer activity of Glycoprotein group one. The results of Trypan Blue coloration suggest that, after incubated with Glycoprotein group one for 24 and 48h with the concentration of not more than 9.829μg/ml, the quantity of live cells is obviously reduced. The results of MTT detection suggest that the Glycoprotein group one can restrain the cell activity of Eca109 under 9.829μg/ml and 4.915μg/ml after 24h; while Hela is the six concentrations between 9.829μg/ml and 9.829ng/ml after 48h.We designed three experiments to find out how the Glycoprotein group one effects on the cells. The results of Annexin V-FITC/PI double fluorescence coloration show that after incubated with the Glycoprotein for 24h, two kinds of cancer cells appear to be the phenomena of apoptosis; for 48h, appear to be the phenomena of cell death. The results of DNA Ladder electrophoresis sustain the conclusion above. According to the result of cell cycle analysis with Flow Cytometer, Glycoprotein can induce Eca109 to be apoptosis after 24h. After 48h, Glycoprotein can reduce the synthesis of DNA in both Eca109 and Hela.
