| Eupolyphaga sinensis was an important resource insect and has been used as traditional Chinese medicine for more than one thousand years.However,the modern pharmacology for E.sinensis was not clear yet.The effects of ethanol extract E.sinensis(EEES)on sport performance,activities and expression levels of antioxidant enzymes in skeletal muscle and liver of exercised rat were analyzed in this study.The EEES were supplemented to rats that were given swimming training at three dose levels 0.5 g·kg-1,1.0 g·kg-1 and 1.5 g·kg-1.The exhaustive swimming time,activities and encoded genes expression levels of superoxide dismutase(SOD),catalase(CAT)and glutathione peroxidase(GST)in skeletal muscle and liver were measured after eight weeks training.The major results showed below:1.The exhaustive swimming time was longer for EEES supplemented rat groups and it was 28.6% longer for 1.5 g·kg-1 supplementation than non-supplemented training rats(P < 0.05),and the exhaustive swimming time was consistent with the supplementation of EEES.The results suggest that EEES is a good candidate for improvement on exercise performance.2.The activities of SOD,CAT and GST were all increased significantly in skeletal muscle and liver among EEES supplemented training rats,compared to non-supplemented training rats: The activities of SOD,CAT and GST in skeletal muscle and liver were increased significantly compared to non-supplemented training rats at 0.5 g·kg-1 supplementation level(P < 0.05);The activities of these three enzymes in skeletal muscle and liver were increased very significantly compared to non-supplemented training rats at 1.0 g·kg-1 and 1.5 g·kg-1 supplementation level(P < 0.01),and the enzymes activity was consistent with the supplementation of EEES.The results suggest that EEES increasing antioxidant enzymes activities in skeletal muscle and liver of training rats.3.The level of gene expression of SOD,CAT and GST were all increased significantly in skeletal muscle and liver among EEES supplemented training rats,compared to non-supplemented training rats: The level of gene expression of SOD,CAT and GST in skeletal muscle and liver were increased significantly compared to non-supplemented training rats at 0.5 g·kg-1 supplementation level(P < 0.05);The level of gene expression of these three enzymes in skeletal muscle and liver were increased very significantly compared to non-supplemented training rats at 1.5 g·kg-1 supplementation level(P < 0.01),and the level of gene expression was consistent with the supplementation of EEES.The results suggest that EEES increased the level of gene expression of antioxidant enzymes in skeletal muscle and liver of exercised rats.In summary,EEES had good antioxidant effect to rats.And the possible mechanism was that the expression levels of encoded genes of antioxidant enzymes increased resulting in an improvement on performance of exercised rats. |
