Effect Of Varicocele On Expression Of Apoptosis Gene Bcl-2 And Bax In Germ Cell Of Adolescent Rats

Background and Objective: Varicocele is a common cause of male infertility, but the mechanism of impairment in testis was not very clear. The past study is mainly about the influence of C02, NO and oxygen free radical in spermatic venous blood to testis,but the report of varicocele on the change of apoptosis gene in germ cell was little. Especially, there is not any report of varicocele on the change of apoptosis gene in germ cell with the duration of varicocele model. So we study on the expression of apoptosis gene Bcl-2 and Bax in germ cell and testis histology in the different time of experimental varicocele rats, to explore the mechanism of infertility by varicocele, and provide experimental evidence to the choice of operative time in varicocele.Materials and methods:90 male SD rats(7weeks) were divided into 2 groups randomly: varicocele group (A group) and control group (B group), A group comprised 60 rats and B group comprised 30 rats. In the A group, we created the model by partial ligation of the left renal vein. According to different time of model, A group was divided into 3 groups(every group had 20 rats). Al group:the rats were killed 2 weeks after the model created, A2 group:the rats were killed 4 weeks after the model created, A3 group:the rats were killed 8 weeks after the model created. In the B group, the left renal vein was exposed without interference with it. According to different time of model, B group was divided into 3 groups(every group had 10 rats). Bl group:the rats were killed 2 weeks after the model created, B2 group:the rats were killed 4 weeks after the model created, B3 group:the rats were killed 8 weeks after the model created. When the model time is up, we did as follows: 1. measure testicular vein diameter. 2. weight the testis. 3. detect the expression of Bcl-2 and Bax in germ cell by immunohistochemical method. 4. observe testis ultrastructure by electron microscopy.Results:1.
