The Effects Of Dietary-induced Obesity On The Fas/Fas-L Apoptosis Pathway Of Testicular Germ Cell Apoptosis In Pubertal Rats

Objective: Explore the role of nutritional obesity,which effects on adolescent malerats testicular germ cell apoptosis pathway of Fas / Fas-L system.Method: Select 24, animal rating of SPF Wistar healthy male rats which age are of 3weeks , randomizedly divided into two groups, namely the control and experimentalgroups, each group have 12 rats. Control mice fed on normal diet; the hyperlipidemiagroup (n=12) were given high-fat and high-calorie diet. The body weight of rats weremeasured in ten weekend, the rats were sacrificed, and then removed the testiclesafter chloral hydrate anesthesia. The correlated indicator for all rats in the peripheralblood serum lipids were detected by automatic biochemical analyzer (ACA) andcollected data,such as TG(triglycerides), TC (total cholesterol), HDL-C (high densitylipoprotein cholesterol) and LDL-C(low density lipoprotein cholesterol). Record thepathological changes of testicular tissues observed under the microscope. Theapoptosis of testis cell was detected by TUNEL staining. The expression of Fas、Fas-Land Caspase-3 protein in testis was detected by immunohistochemistry.Result: High fat group compared with control group, the TC, TG and LDL-C andHDL-C was significantly increased, the difference was significant (P<0.05). Germcell apoptotic index of high-fat group was significantly increased than the controlgroup (P＜0.01). Apoptotic cells are spermatogonia and spermatocytes. Theexpression of Fas、Fas-L and Caspase-3 protein in hyperlipidemia group wasdefinitely higher than the control group(P＜0.01).Conclusion: Nutritional obesity factor induced rat testicular germ cell apoptosis isincreased, its mechanism may be related to increased expression of apoptotic factorsFas, Fas-L and Caspase-3 protein.
