Objective This study was conducted (1).to establish stable experimental model of rat orthotopic liver transplantation(ROLT), summarize operative skills and techniques, so as to make adequate preparations for the next research. (2) to observe the relations between the NK1.1T cells'homing to liver and Kupffer cells and liver sinusoidal endothelial cells,to study the relations between the NK1.1T cells'homing to liver and the mutual action between the ICAM-1 and the LFA-1. Through these researchs we want to clarify the mechanisms of NK1.1T cells'homing to liver after liver transplantation.Methods (1) The animal model of orthotopic liver transplantation in rat was performed with two-cuff technique, in which suprahepatic vena cava (SHVC) was anastomosed with continuous suture, portal vein (PV) and infrahepatic vena cava (IHVC) were anastomosed with cuff method, and the bile duct was reconstructed by interior stent.
