PARTâ… EFFECTS OF MAGNIFICATION AND EMBEDDING ON THE MORPHOMETRIC STUDY OF TESTICULAR STRUCTURESBackground and Objective:In morphometric studies, we usually use tissue sections to estimate the quantitative morphological characteristics of structures, based on measurements of images observed on sections according to stereological principles. As sections are affected by many factors such as sampling of materials, fixation, dehydration, embedding, sectioning, mounting and staining of sections, and microscopic resolution or magnification, different results may be obtained under different conditions.. This is a basic and important question, but systemic studies or reviews on it are lacking, probably because it is affected by so many factors (including measurement methods). The authors' laboratory once compared the effects of embeddings with the homemade paraffin and epon resin on the morphometric study of testicular structures, and, recently, also compared effects of embeddings with the internationally commonly-used Leica paraffin and methacrylate resin and magnifications on the morphometric study of renal structures.
