Abjective:Invitro culture and induce dendritic cells separated from patients infected with CHB invitro.Measure the concentration of GSH-Px and MDA in DC,concentration of IL-12 in DC culture medium and DC's competence of stimulate multiplication of Tlymphocyte from the same person。To observe antivirus ability of DC invitro,discuss DC function abnormity in patients infected with CHB and seek new approach ameliorating DC function.Method:(1)Invitro culture PBMC separated from patients infected with CHB and the health,using substrate AIM-V and GM-CSF,rhIL-4,rhIFN-α. (2)Collect peripheral blood of 20 sufferer and 8 health people .group them into three groups:treat group,cultured with Sodium Selenite ;hepatitis group,cultured without Sodium Selenite;control group,health control.(3),Mensurate CD80,CD86 expression on DC's surface with Flow cytometry and mensurate HLA-DR expression on DC's surface with immunochemistry.Use Wright ' s staim coloration,transmision electron microscope and scanning electron microscope to identify CD's configuration.
