Electrochemotherapy is an efficient local therapy for cancers,which combines chemotherapeutic agent and short, high voltage electric pulses. It can enhance chemotherapeutic agent to enter into the cells, thereby reducing the dose needed and decreasing the side effects. Steep pulsed electric fields (SPEFs) were developed electrochemotherapy in recent years, it does not rely on chemotherapeutic agent but on the electric fields which lead to irreversible electrical breakdown (IREB) in tissues.Objective The aim of the present study was to investigate the dosage of SPEFs. To study injuring effects of different metrology of SPEFs on fresh excised calf liver and rabbit liver. A method to monitor the effect of SPEFs was also investigated.Materials and methods Fresh excised calf liver tissues were exposed to steep pulse electric field with serial voltage, duration and frequency, and then the necrosis tissue diameter was measured. B-mode imaging was used to visualize the gray change before and after steep pulses. TTC and histological examination were used to determine the injuring effect.
