Objective: To establish a liver cancer and its angiogenesis model in order to study the biology of tumor and their angiogenic in chick CAM.Method: Human liver cancer cell of SMMC-7721 were planted on 8-day chick CAM with 1× 106-4×107 per egg,. The cell densityes inducing tumor tissue growth and its angiogenesis were tested.After planting of 6×106 per egg, the tissue growth and its angiogenesis were observed daily.Result:The growth of tumor tissue and its and its angiogenesis on CAM were related to the cell density planted.The best cell density planted for tissue growth start and its angiogenesis was 4× 106/egg and 6× 106/egg, respectively (P<0. 05). The best cell density for maximal tumor tissue growth and its angiogenesis was 1×107/egg, The cell densityes recommened for both tumor and its angiogenesis was between 6 × 106-1×107/egg. The angiogenesis reaction and tumor growth were observed 2-3 days after planting,The angiogenesis occurred rapidly between 3th - 4th day (P<0. 05) and attained to the peak on day 5. The groeth of tumor was 1 or 2 days later than angiogenesis.Conclusion: 6× 106-1×107per egg was the situable cell density for the establishment of model in CAM with Human liver cancer cell line SMMC-7721. the angiogenesis and tissue grow were obviously on day5-6th. Planted with this model is a useful tool to study biological characterize of hepatocellular carcinoma and for drug discovery, it is a relative rapid and easy model to study hepatocellular carcinoma and antiangiogenesis dryg screening.
