Objective:To observe the effects of promethazine on the activity of mitochondrial enzyme,the concentration of mitochondrial calcium and mitochondrial transmembrane potential (Δ Ψm). Methods:72 male SD rats were randomly allocated into four groups(n=18):①normal control group; ②brain ischemia group; ③promethazine 1 group (P1 group); ④promethazine 2 group(P2 group). According to brain ischemia-reperfusion time(1h, 6h, 24h),each group was again divided into three timepoint-subgroups (n=6): 1h(I10minR60min),6h(I10minR360min),24h(I10minR360min). P1 group and P2 group were intraperitoneal injected with promethazine 10mg/kg and 20mg/kg on 1h before operation respectively while normal control group and brain ischemia group were given equivalent volume saline. Animals were killed on 1h, 6h, 24h after operation respectively, and cerebral mitochondria were isolated. Protein concentration of the mitochondrial suspension was measured. Purity and lives of mitochondria were determined by the succinate dehydrogenase(SDH) activity. The concentration of mitochondrial calcium and Δ Ψm were measured. Results: ①The SDH activity of three time points in brain ischemia group significantly decreased comparing with those of normal control group(P <0.01).
