Objective1. To determine the heating characteristics and impact factors on heat productions of the magnetic fluid in alternating magnetic field.2. To assess the therapeutic of magnetic fluid hyperthermia on Lewis lung carcinoma.Methods1. Different weigh (10mg, 15mg, 20mg) of magnetic fluid were prepared in vitro,and were heated for 30 minutes within an alternating magnetic field (frequency 100-250kHz, amplitude 7-15kA/m), Influences of other parameters on magnetic fluids were achieved by adjusting intensity of magnetic field, respectively.2. 80 C57BL/6 mice were inoculated with Lewis lung carcinoma cells. When the tumor grew to 0.8cm±0.1cm in diameter,all the mice were divided into four groups :control group,magnetic group,magnetic fluid group and hyperthermia group.Magnetic fluids were directly injected at the center of tumors in hyperthermia group,then subjected to magnetic field for 30 min.After hyperthermia, the tumor tissues were removed and histologically analyzed to access the thermal destruction after treatment.The change of tumor volume were observed.And the tumor growth inhibitory rate was caculated .Flow cytometry was used to analyze apoptosis rate and the change of cell cycle.Leukemia cell counts were measured.Survival were compared among the groups .
