OBJECTIVE:To establish the colonic liver metastases model in BALB/c mice. Observed in vitro magnetic field strength of the relationship with the local temperature rise in colorectal cancer metastasis model, Observed magnetic hyperthermia killing effect on tumor tissue and adjacent tissue thermal damage effect of the concentration of the magnetic fluid and magnetic field strength of the dose-effect relationship; Research Study magnetic fluid embolization of the tiny blood vessels in the injection of local, as well as the tiny blood vessels of the body tissues without embolic side effects.Provide some data for future clinical treatment of colon cancer liver metastases.METHOD:BALB/c mice were randomly divided into3groups with injection of0.2ml CT-26CRC cells (1×107ml) by intra-abdominal spleen transplanting (Spleen removal group and spleen reservation group),rectum transplanting respectively. The conditions of all the mice were observed, and the survival time and liver metastases rate were recorded.Department of animal models:Take1104-week-old female Kunming mice, select the spleen injection and spleen reservation established mouse tumor model. The experiment is divided into5groups:Magnetic field strength and the local temperature group:When hyperthermia for20min, and the concentration of the magnetic fluid7.8mg/ml,Observed in the magnetic field strength, respectively,320A,420A,520A,The tumor local temperature changes,take tissue by HE staining in tumor cell killing effect of different magnetic field;Hyperthermia and local temperature Group:When the magnetic field strength is520A, the concentration of the magnetic fluid7.8mg/ml,The observed hyperthermia time for20min,30min,40min,The tumor temperature changes,take tissue by HE staining in tumor cell killing effect of different magnetic field;Magnetic fluid concentration and local temperature Group:When the magnetic field strength520A, hyperthermia20min,Observed in the magnetic fluid concentration of3.9mg/ml,7.8mg/ml,15.6mg/ml,The tumor temperature changes of tissue by HE staining in tumor cell killing effect of different magnetic field;Adjacent organs thermal injury group:Take the best experimental results of the magnetic field strength, hyperthermia time, the concentration of magnetic fluid,Mice after hyperthermia by HE staining, HSP70immunohistochemistry to study observed adjacent organs of mice lung, kidney, heart and thermal damage.Magnetic fluid embolism group was divided into4groups:Negative control group, the control group of the magnetic fluid, the magnetic field control group, the magnetic fluid hyperthermia group,Each group tumor tissue and liver tissue, HE staining observed contrast magnetic fluid embolism tiny blood vessels.RESULT:Modeling Experimental:The colonic carcinoma liver metastasis model was established by all of three ways. Spleen removal group, the incidence rates of liver metastases was100%, survival time was (28土5) days, there were lung, stomach, diaphragm to transfer; Spleen reservation group, the incidence rates of liver metastases was95%, survival time was (30土5) days, there were lung, stomach, diaphragm to transfer; Rectum transplanting group, the incidence rates of liver metastases was30%, survival time was (20土5) days, no lung, stomach, diaphragm metastases;Magnetic field strength and the local temperature group come to the best electric field strength520A,Hyperthermia and local temperature group drew the best role for20min,MHD concentration and local temperature group derived the best magnetic fluid concentration of7.8mg/ml;Adjacent organs thermal injury group after treatment adjacent organs of lung, kidney, heart tissue HE staining heat damage, no heat damage effects.Magnetic fluid embolism group, negative control group, no liver tumor thrombosis;The blood vessels in the the MHD control group liver tumors visible to the magnetic fluid accumulation, but not the formation of embolism;The magnetic field control group, no liver tumor vascular thrombosis; magnetic fluid hyperthermia liver tumor blood vessels visible micro-thrombosis.The immunohistochemistry Tips:hyperthermia and heat shock protein70(HSP70) in tumor cell cytoplasm were positive, compared with control rats;CONCLUSION:Modeling Experimental Spleen reservation group and Spleen removal group had higher rates of liver metastases,longer survival time than rectum transplanting group. The modeling is more stable, better able to complete the evaluation of the indicators in the observation period. When the concentration of the magnetic fluid hyperthermia same time, the tumor localized temperature increases with the enhancement of the magnetic field strength, time, the magnetic field strength is520A. When the magnetic field strength, magnetic fluid concentration is constant, the tumor temperature increase with the growth of hyperthermia time, the best hyperthermia for20min; When the magnetic field strength, hyperthermia same time, tumor localized temperature increases with the increase of the concentration of magnetic fluid, the the preferred magnetic fluid concentration of7.8mg/ml. After tumor injection of magnetic fluid hyperthermia in the external alternating magnetic field, can make the blood vessels within the tumor bleeding,reongestion, congestion, the occurrence of vascular thrombosis.
