| To investigate the mechanism of acute lung injury induced by pancreatitis associated ascetic fluid (PAAF) and explore the octreotide's protective effects on it, so as to provide the oretical principle of the treatment of acute hemorrhagic necrotic pancreatitis (AHNP) . [Methods] : Forty healthy Sprague-Dawwley (SD) rats, fifty percent of witch were male , were made into acute hemorrhage necrosis pancreatitis (AHNP) models , and eight of them were choosed randomly as AHNP group whose lungs and pancreas were observed and their AmYL in serum were measured. Forty eight healthy Sprague-Dawwley (SD) rats, male or female, were randomly divided into three groups: group A served as the control group and 8ml saline (NS) was injected into the peritoneal cavity; group B as the model group, 8ml PAAF was injected ; group C as the treatment group,1h before PAAF injected and then 2h 5h, 8h, 11h after 8ml , was administrated octreotide subcutaneously with the dose of 4ug/Kg and 20ug/Kg ; respectively, NS replaced octreotide in group A and group B with the same volume. After peritoneal cavity injection, the rats were put to death in two baches at 7h and 12h. Level of AmYL, LPS, TNF-a and WBC in blood, Respiratory frequency, PaO2 , et al , were measured. [Results] : the level of TNF-a, LPS , R, AmYL in serum and WBC in group B were significantly higher than those in group A (P<0. 01-0. 05) ; The level of PaO2 in group B was significantly lower than that in group A (P<0. 01) . After injection of octreotide, the level of TNF-a, LPS , R, AmYL in serum and WBC in group C was lower than group B (P<0. 01-0. 05) , but the indexes of blood gas analysis and the level of lung wight/dry showed no obvious change in group C(P>0. 05vs B). The lung tissue showed obvious pathologic changes in group B, but, after octreotide given, the lung tissue has no obvious pathologic changes in group C. [Conclusion] : PAAF can induce acute lung injury in healthy SD rats, and the animal model of acute lung injury induced by PAAF was established successfully; octreotide has no... |
