[Objective] To observe the effect of deep hypothermic circulatory arrest (DHCA) on expression of HIF-la(hypoxia—inducible factor la), Bcl-2 and Bax mRNA in cortex neuron of rats.[ Methods ] Eleven SD rats were divided randomly into three groups : deep hypothermic circulatory arrest (DHCA) group, normal temperature circulatory arrest (NTCA) group and normal animal control (NC) group. The animal model of closed-chest DHCA was established in rats by means of external cannulation. In DHCA group, circulatory arrest were performed in the deep hypothermia (18°C); in NTCA group, circulatory arrest were performed in normal temperature and in NC group without circulatory arrest and deep hypothermia. 5 mins later cerebral cortexes were removed and then dissociated ,measured HIF-1a, Bcl-2 and Bax mRNA by Real-time PCR. Data were analyzed by software SPSS13.0.[Results] The level of HIF-la and Bax mRNA expression of DHCA group and NTCA group was much higher than of NC group(P<0.05). Compared with NTCA group, HIF-la expression of DHCA group was much higher (P<0.05) but Bax mRNA expression was lower (P<0.05). However, the level of Bcl-2 mRNA expression in all groups were similar to each other(P>0.05).[Conclusion] Ischemia can induce the expression of HIF-la and Bax mRNA while deep hypothermia increase HIF-la expression much more and on the other hand deep hypothermia decrease Bax mRNA expression.
