Objective Observe the inhibting effectt of TNF-α on hypertrophic scar and study the mechanism. Provide the theoretical and experimental basis for curing hypertrophic scar.Methods 15 of big ears Japan white rabbits were used to establish the model of hypertrophic scar of rabbit ears. There are 5 hypertrophic scar pieces on each rabbit ear. Divide the hypertrophic scar pieces into 5 groups. A,B, C, D groups were injected TNF-α 0.1ml( consistencycorder: 10ng/0.1ml,50ng/0.1ml, 100ng/0.1ml, 500ng/0.1ml) and E groups with NS 0.1ml. Seven days later, the influence of injecting TNF-α was researched by histomorphology. The immunohistochemistry staining technique was used to detect the expression of caspase-3 protain.Results The staining of HE of A, B, C, D groups showed that the volume of hypertrophic scar of rabbit ears were lower than the control groups, the number of fibroblasts of scars was obviously less and the arrogements of collagen become order. The inhibiting effect of TNF-α on model of hypertrophic scar of rabbit were stronger with the increase of TNF-α consistency.Conclusion Those experimental results showed that TNF-α with high consistency is able to inhibit the proliferation process of hypertrophic scar of rabbit ears and to remarkably decrease the degree of fibrosis to scars. The apoptosis of fibroblasts are important process during the course of inhibit the proliferation process of hypertrophic scar of rabbit ears. The caspase-3 protein maybe related to the mechanism.
