Subject To research the influence of different concentrations of kinetin on the proliferation of human fibroblasts in vitro and study the effect of locally and externally use of the kinetin on the rabbit ear hypertrophic scar models.Methods(1)Extract the human fibroblasts from the scar tissue which come from surgery.Prepare the culture medium containing different concentrations kinetin to cultivate people fibroblasts.Through the CCK-8 detect the proliferation of human fibroblasts and flow cytometry instrument detect the growth cycle of human fibroblasts.(2)Making the rabbit ear hypertrophic scar models:12 Japanese big ear rabbits were chosen to make 6 round wound which 1 cm in diameter on the side of one ear abdomen by remove the full-thickness skin and perichondrium.We apply the kinetin on the rabbit ear hypertrophic scar models after 21 days injury when wound scab off.We set every white rabbit's right ear hyperplastic scars as experimental group that daubed mass concentration of 80 mg / kg of kinetin vaseline ointment twice a day.We set every white rabbit's left ear hyperplastic scars as comparison that daubed isodose vaseline ointment twice a day.Gathering the rabbit ear photos after 1 week,2 weeks,3 weeks,4 weeks and 5 weeks.The observed indicators are scar color,texture and bulge.We measure the scar diameter and make scar section for HE staining and Masson staining.We compare the difference of scar tissue elements between the two groups and calculate the scar proliferation index.Result(1)When the concentration of kinetin ?20 mg/l,they play a promotional role in proliferation of cultured human fibroblasts in vitro.When the concentration of kinetin is 20 mg/l,the promotion effect is strongest.When the concentration of kinetin?40 mg/l,they play a inhibition role in proliferation of cultured human fibroblasts in vitro.With the increased concentration of kinetin the inhibition is enhanced.Therefore,the concentration range can be as kinetin local external use of rabbit ear hyperplastic scar effect research.(2)On the surface,compared with the control group,the experimental group rabbit ear scars are lighter in color,more soft in quality of the material and more flat.The diameter of the scars have no significant difference between two groups.Observation from the perspective of histological,compared with the control group,the experimental group rabbit ear scars are less in number of fibroblasts and collagen content.The dermis are obvious thin.Collagen bundles is slim and loose and arrange more rules.The infiltration of inflammatory cells have a significantly reduce.The microvasculars are decline in the number.The scar proliferation index is decreased significantly.Conclusion Kinetin can inhibit the growth of fibroblasts to reduce the rabbit ear scar hyperplasia.By local external use the kinetin to reduce the scar hyperplasia have a Significant effect.Kinetin have a considerable application prospect in treatment and prevention of hypertrophic scar.This experiment has provided a new train of thought for looking for effective prevention and treatment of hyperplastic scar by drugs.
