Objective To investigate the effect of Hypericum perforatum(HP) on the acute injury of rat pulmonary microvascular endothelial cells(rPMVEC) induced by paraquat (PQ) and its probable mechanism. Methods RPMVEC would be cultured in vitro, using tissue mass. The rPMVEC were divided into 4 groups: PQ group, HP protecting group(3 concentrations), HP control group(3 concentrations) and blank control group. After adding 0.1mmol/l PQ to both the PQ group and the HP protecting group for 0.5h, the time started, and then, the medium and HP were given to the PQ group and HP protecting group respectively. MTT was measured on different time spots (3, 12, 24, 48 and 72h) to test the changes of the growth inhibit rate. Meanwhile, the activity of total SOD and the contents of MDA in supernatant were detected by spectrophotometry. Three different concentrations of HP(250mg/l, 10mg/l and 1mg/l) were detected respectively, to compare the different effects of different concentration of HP on the acute...
