ObjectiveThis study aimed to observe the antioxygent effect of ambroxol hydrochloride and reduced glutathione hormone in the acute lung injury model of rat induced by paraquat by detecting the malondialdehyde(MDA) content and superoxide dismutase(SOD) activity in lung tissue.Look for a ideal medicine to cure the provent acute lung injury induced by paraquat. Methods1. The acute lung injury model of rat was built by paraquat i.p. Ninety healthy SD rats were randomly divided into five groups, eighteen animals each.Group A: reduced glutathione hormone group; Group B: ambroxol hydrochloride group; Group C: ambroxol hydrochloride and reduced glutathione hormone group; Group D:postive control group;Group E:negetive control group. Every mouse of group A, group B, group C, group D was given paraquat at the dose of 20 mg/kg ip , Every mouse of group A was given reduced glutathione hormone 200mg ip; every mouse of group B was given ambroxol hydrochloride 50mg/kg/day ip; every mouse of group C was given ambroxol hydrochloride 50mg/kg/day ip and reduced glutathione hormone 200mg ip; every mouse of group D was given 0.9%NaCl 2ml/day ip.Each group was divided into three subgroups (n=6); subgroup a:24h later;subgroup b:48h later;subgroup c:24h later.The pathologic changes of lungs were measured.The malondialdehyde (MDA) content and superoxide dismutase(SOD) activity in lung tissue was detected by colorimetry method.Results1.Pathologic changes:Group D: l,2days after the mice were given paraquat: the lungs were larger than before, alveolar wall bleeding, edema of lung. Compared with group D the congestion and bleeding alveolar wall of group A, group B and group C were lessened. There is no significant difference between groups of A, B and C.2.In each groups given paraquat the activation of SOD was significantly increased and the contents of MDA significantly decreased compared with group E(P <0.01). Compared with group D, the activation of SOD of group A, group B, group C were significantly increased and the contents of MDA significantly decreased. There is no significant difference between groups of A, B and C(P>0.05).Conclusions:1.Ambroxol hydrochloride or reduced glutathione hormone has the antioxygent effect on the acute lung injury model of rat induced by paraquat.2.The combination of ambroxol hydrochloride and reduced glutathione hormone have the antioxygent effect on the acute lung injury model of rat induced by paraquat.But the effect it is not better than single use.
