Objective: After samples being pretreated by solid-phase extraction (SPE) technique, two analytical methods of gas chromatography-flame ionization detection (GC-FID) and gas chromatography-mass spectrometry (GC-MS) for determination of nicotine and its metabolite cotinine in human's urine were proposed. Based on the laboratory quality control, 607 samples have been detemined by GC-FID. They provided the reliable methods and data for the tobacco control and smoking intervention in China.Methods: The influence of parameters on the pretreatment and chromatographic condition for nicotine and cotinine was investigated and the procedure carried out as follows:1. The samples were separated into three groups of non-smokers, passive smokers and active smokers by the basic investigation. Nicotine and cotinine were enriched and purified by solid-phase extraction (SPE) using C18 column. After being adjusted to pH7.5 with phosphate buffer solution (PBS) , the samples was washed with the ammonia solution and eluted with methanol-acetric acid solution by the static elution. The elution was evaporated with nitrogen and analyzed by FID-GC. The contents of nicotine and cotinine in samples were quantited with calibration curve.2.
