Backgroud: Mesenchymal stem cells (MSCs) are nonhematopoietic progenitor cells of the bone marrow stromal cells. MSCs have the ability of differentiating into various tissues of mesoderm sources. MSCs can also regulate the function of the immune system. In vivo studies show that allogeneic stem-cell transplantation can reduce the incidence and severity of acute and chronic graft-versus-host disease (GVHD) and prolong skin graft survival. There are growing data showing that MSCs have immune regulation functions, especially the inhibitory effects on lymphocyte proliferation. However, the mechanisms of the immunosuppression are still obscure.Object: To study effects of mesenchymal stem cells (MSCs) on lymphocyte proliferation and cell apoptosis and possible mechanisms.Method: MSCs were isolated and cultured from BALB/c mice routinely.With 3 passages, MSCs showed typical appearance and immunophenotype(CD29+, CD44+, CD105+, CD34- and CD45-) and multipotential.
