| BCG injection is one of the most efficiency way to prevent children from tuberculosis all over the world. The mycobacterium such as M.phlei, M.vaccae, are widely used for vaccines and adjuvants. Mycobacterium agents were restricted to use in clinic because some components of mycobacterium have no immune activity or immune inhibition, and lead to positive in tuberculin test.Using biochemistric method, the quantity of thalli protein and peptide extracted from mycobacterium was detected in this report. According to animal experiment, the specific and non-specific immune effects induced by thalli protein and peptide were studied. A new type with low toxic and high immune effect adjuvant was identified and the best dosage of thalli protein and peptide was confirmed.By means of lowry's detection and Tricine-SDS-PAGE, the thalli protein and peptide extracted from mycobacterium in low temperature was analyzed. The results showed that the quantity and components of the thalli protein was steady and no significant differences between different extracting times. The molecular weights of the multi-components of thalli proteins were from 66 kD to 6.5 kD.The quantity of the drug given to broilers was based on the human dosage calculated from the area proportion between broiler and human. 210 broilers were randomly divided into seven groups such as control, high dosage of thalli protein (0.9mg per broiler) , modarate dosage of thalli protein (0.45mg per broiler) , low dosage of thalli protein(0.225mg per broiler) , peptide (0.3mg per broiler) , mycobacterium (0.3g per broiler) injecting of thalli protein (0.45mg per broiler) , respectively. All thalli proteins were given orally except injecting group. All broilers were killed at 7d, 14d, 21d and 28d after given drugs.
