Objectives:To use nano-aluminum hydroxide adjuvant(NAHA)assist to obtain rapid advance and higher of antibody for highly pathogenic avian influenza virus(HPAIV H5N1).NAHA reduce the oil adjuvant current widespread and increase carcass quality of broilers.Methods:1.The particle of NAHA was prepared by microemulsion.The third and fourth cycle of the product in high-pressure homogenization process were selected,their particle size was measured by Zetasizer 3000HS.2.Study on sterilization of NAHA.Turbimetric Assay(TA),the pH and the size of Nanometer-Particles(NPs)were applied to this comparison for before and after autoclaving(121℃30min).3.Vaccine with HPAIV(H5N1)and two Nanometer-Particles(NPs)vaccinated broiler.Oil adjuvant,two NPs adjuvant,that is oil-group,NP80-Group,NP219-group,the right chest muscle side is the experiment part and control-group.Wing vein blood was collected in 14 days old,17 days old, 19 days old,21 days old,24 days old,28 days old,31 days old,35 days old,42 days old,49 days old.Antibody level was measured with micro-hemagglutination inhibition test.In 21 days old,28 days old,35 days old,42 days old,heart blood was collected and separated lymphocytes,The lymphocytes was determined on EAC and MTT.4.NP80-Group,NP219-group and oil-group(the left side is control)was evaluated with contrast of oculo-view on 17 days old and 21 days old,pathologic histology and the value MPO on 21 days old,35 days old,49 days old.Results:1.Two mean diameter of 80nm and 219 nm NAHA was procured..They was measured by Zetasizer 3000HS.2.The results revealed the pH was slightly decreased during the autoclaving,but there was not significant deviation about turbimetric change or diametric change of the NPs.NAHA can be sterilized by autoclaving during the production of bacterin.3.The results revealed NP80-Group and NP219-group(21-day old)procured antibody the response 3 days earlier than oil-group(24-day old).Antibody titer of NP80-Group and NP219-group was not significant deviation.Antibody of persistence of oil-group is longer.21 days old,28 days old,35 days old,42 days old,Rosette forming rate(EAC)and transformation rate of lymphocytes(MTT)of NP groups,oil group and the control group did not significant deviation(P<0.05)4.The results revealed NP80-Group and NP219-group were more excitation(after vaccinated 3 days).Muscle is red and oedema,whereas oil-group less and malabsorption,gradually to form yellow trail.H.E staining of the part muscular tissue revealed that inflammatory cells assembled phlegmasia focus stronger early than later.Oil-group still revealed phlegmasia cells focus on 49 days old,whereas two NAHA groups are few on the day.The level of MPO illustrated the value of MPO is high early,opposite later.Oil-group is highest at 35 day,Four groups are all slight at 49 days old.Two NAHA groups are not significant deviation.Conclusions:Two NP adjuvants was prepared by microemulsion.They can be sterilized by autoclaving(121℃,30min).After the sterilization treatment the turbidity and particle size is no significant impact.The pH value decreased,but no fundamental change of electrical.Vaccine with NAHA is better than oil adjuvant on immunology.Vaccine with NAHA is smaller for the impact carcass quality than oil adjuvant on oculo-view test,histopathology and MPO value.HPAI has spreaded in Asia,Africa,Europe and other regions worldwide.The epidemic was caused poultry,wild birds and human deaths.Effective immunization is one of the important measures for the prevention and control of HPAI outbreaks and spread.Vaccine with whole inactivated virus and oil adjuvant is utility for the disease.The antibody of level of HPAI HA is evaluate index after immunization.As we all know,immune adjuvant increase immunization levels.Oil adjuvant only use for animal vaccines.Although the study of other immune adjuvant is very broad,but not use widespread.Aluminum hydroxide was widely recognized by FDA et al.Aluminum hydroxide enhanced the antibody of some antigens in the immune response for more than 80 years,and safety.With time gone, Aluminum hydroxide revealed some faults.Such as:slow antibody,antibody levels are not high, injection site of the local strong stimulus,and new vaccines(subunit vaccine,the DNA vaccine)in response to the further development,too.The APCs swallowed up antigens by selective differences after the antigen into the body et al.In conclusion,although aluminum adjuvants have been used in vaccines for more 80 years, why to improve humoral-mediated immunity(HI),how to improve HI and how to produce more antigen to induce strong cellular immunity is still a key problem to be studied in the future.We are sure that the nanoparticulated alum adjuvant will have a bright future if further studies are performed.
