Objective: to observe and compare the expression of inducible nitric oxide synthase and peroxynitrite anion and the charactistic of apoptosis related to signal transportation of Caspase-3, which is invovled in the pathogenesis of the biologic response to alumina and ultra-high-molecular-weight polyethylene (UHMWPE) particles in vivo. The purpose was to investigate the mechanism of the periprosthetic osteolysis by wear debris during asepetic loosing. Methods: alumina ceramic and UHMWPE wear debris were less than 10.0. m in diameter, which are produced in vitro. using a murine air pouch model of inflammation, Kun Min mice were divided into 3 groups in the present study. The air pouches were injected with 3ml of suspension containing 1×10~8/ml alumina particles in group A, while UHMWPE particles in group B, control pouches received 3ml of sterile PBS alone. All animals were sacrificed respectively at 3,7, 14 d after the injection into the pouch. Histological changes and cell counting were assessed by light microscope, TUNEL assays were conducted to quantify apoptotic cells and immunohistochemistry method was applied to detect the expression of Caspase-3,iNOS,NT in the pouch membrane.Results: increased cell infiltration,TUNEL-positive cells and Caspase-3,iNOS, NT immunoreactivity expression were detected in group A and B compared to control; cell number and the apoptotic index and expression of Caspase-3,iNOS, NT in group A and B was significantly higher than that in control group (P<0.01); cell number in group B was significantly higher than that in group A at all time points (P<0.01); expression of iNOS,NT in group B was higher than that in group A at Day 7(P<0.01); the apoptotic index and expression of Caspase-3 in group B was higher than that in group A at Day 7,14 (P<0.01); the apoptotic index correlated with the expression level of Caspase-3(r =0.752, P<0.05).
