Objective By using tissue chip to detect the difference of expression and correlation and significance among CD117, CD34, SMA, S-100, ERK1, p16 and Ki67 in GISTs, hoping to explain the mechanism of GISTs and the role of ERK1 in inducing cells proliferation and differentiation which might provide some basic datas for clinic study.Methods 1. To make a tissue chip by using collected samples of 40 GIST and 40 normal control group:①to design the tissue chip;②to dig holes on the blank paraffin block using the design, and the diameter of holes is the same size with tissue we take from paraffin embedded blocks;③watching samples of GISTs' histopathology on HE sections, and mark the point of the place that we are going to dig tissue from; ④by using special digging machine to dig tissue and then transfer it to the accordingly place of the blank paraffin blocks one by one till we finish all the samples and make notes on paper for later watchin and counting; ⑤ the sections were cut at 4μm and stained with hematoxylin-eosin and Maxvision immunostaining; 2. Immunohistochemical staining for the paraffin sections by using polyclonal antibody ERK1 and monoclonal antibodies p16 and Ki67 was performed by Envision two steps technique in 40 GIST group and 40 normal control group and monoclonal antibodies CD117, CD34, SMA and S-100 in 40 GISTs using the same way.Results 1. The expressions of ERK1 and Ki67 were higher in GIST group than that in normal control group, but the expression of p16 was on the contrary in these two groups. And the differences had statistic...
