Percutaneous transluminal coronary angioplasty(PTCA) has become increasingly important in the therapy of coronary artery disease. However, the high restenosis occurrence of 20%~30% in patients limits the applications of this method, but the pathogenesis of restenosis has not yet been fully understood. What has been perhaps underappreciated in the evolving understanding of restenosis from animal model and clinical perspectives is the profound impact that a chronic indwelling stent has on the vascular biological response to coronary intervention, particularly the impact on the inflammatory response. In order to research the pathogenesis of restenosis, peoples set up many kinds of in-vitro ways. Many important information can be foud by organ culture model, it has become into a trend for researching in-stent restenosis by using exvivo model.PURPOSES: (1) To set up the organ culture system so as to provide valuable information for studies of in-stent restenosis after the stent implantation, especially for the evaluation of new stents. (2) To research the drug-coated stent function, if there has the obvious anti-inflammatory function. (3) Inflammatory releasing indicate that the vessel is stimulated by an implanted stent, whether there has the direct relationship between inflammatory releasing and neointimal hyperplasia.METHODS: (1) Preparation of the artery organ-culture system. (2) Preparation of atherosclerotic rabbit model: combined the methods of high-lipid diet and immune injure. (3) Studies of in-stent restenosis: morphometry and inflammatory releasing.RESULTS: (1) Comparing of static culture(without sheer stress),the vessel without a stent maintained the original morphology and normal activity which cultured in the organ culture system. (2) The arterial vessel of 8 week atherosclerotic model was found by microscope. (3) Comparing of stainless steel stent, it appeared less proliferation around the stent with curcumin coated.
