OBJECTIVE The study is designed to demostrate the protective effects of ST-1 against cerebral injury induced by focal ischemia-reperfusion in rats and to explore the possible mechanism by testing the expression of NF-κB and Bcl-2.METHODS Adult healthy rats are random divided into six groups, including sham operation group, ischemia-reperfusion group, nimodipine-pretreatment group and groups of high, medium and low dose of ST-1 pretreatment. The model of focal ischemia-reperfusion is induced by middle cerebral artery occlusion for 2 h followed by reperfusion 22 h in rats which is based on the method advanced by Koizumi. Neurological function deficit scores are evaluated, Infarct size, malondialdehyde (MDA) content and superoxide dismutase(SOD) activity are measured, pathological changes of neurons in hippocampal CA1 region and cortex are observed. The expressions of NF-κB and Bcl-2 are observed through immunohistochemistry.RESULTS The ischemia-reperfusion causes large scale of infarct and evident changes in behavior, biochemical indexes, pathology and in expressions of NF-κB and Bcl-2. Compared with vehicle group ST-1 at dose of 10 mg/kg and 20 mg/kg can reduce the infarct size,MDA content, enhance SOD activity and relieve neurons injury in hippocampal CA1 region in dose dependent way; ST-1 at dose of 20 mg/kg can also improve neurological...
