Objective:We established a different lodas of the rat's exercise model by swimming in order to observe histological features loaded different movement force and expression of corresponding adhesion molecules,expecting which will help for elecidating the mechanism that overload training induced the prejudice of liver and kidney.Methods:Thirty-six Wistar adult male mice was selected randomly and divided into three groups respectively.Control (C) group(n=12) was raised naturally without intervention at all.Generally Swimming Trained (GST) group(n=12) was performed with swimming training for 6 days in a week,and once a day,trained in general movement force.Overload(O) group(n=12) was performed with overloaded swimming training.Then they were killed in the next day after full eight-week training. The tissues of liver and kidney were selected respectivily.Fixing-dehydrating-transparecy-embed-slice were perfomed.Midsagittal setions 5um thick were stained with haematoxylin and eosin.We examined the expression of intercelluar adhesion molecule-1(ICAM-1),P-selectin in hepatic and renal tissues by immunohistochemistry.Results:1.General condition of C and GST was normal.O was sleepy and apt to be frightened.Whose hair abscission and somatotype was emaciated and eyes intnese darkness without light.2.In C rats,the structure of hepatic lobule were integrated,hepatic cables arranged orderly,hepaticsinusoid were eyeable.the structure of renal tubule was normal,no hyperaemia and edema in interstitial growth.Typical histological features were seen in those experiment models.In GST rats,some of hepatic cables were broken,hepatocyte were swellen gently and a few inflammation cells soaked among them.Renal tubles collacated appreciably inordinate and were lightly metamorphic,renal glomerulus were intumescent.As movement force was increased by degrees,pathological changes became more serious hereinbefore in the liver and kidney.3 Immunostaining by anti-ICAM-1and anti-P-selectin positive in renal glomerulus endothelium cells,renal tubule epithelium cells and hepatocytes.The expression of ICAM-1and P-selectin showed a increasing tendency with the increase of training load.Our study indicated that the amount of ICAM-1and P-selectin positive cells in O rats were significantly more than that in C rats.There were no significant difference between GST rats and C rats.Conclusions:1Obvious pathological changes were not seen in rat's liver and kidney after general swimming training.In O rats,we observed serious histological...
