Objective: (1) To isolate the active compound from the extraction of Hydrozotyle sibthorpioides Lam. (2) To prove the inhibitive effect of HSA against carbon tetrachloride (CCl4)-induced liver fibrosis in rats. (3) To study protective effect and involved possible mechanisms of HSA. Methods: (1) Isolation of active compound: Subject the fraction to column chromatography on silica gel, and two compounds were obtained.Seive the active cmpound by chronic liver injury experiment. (2) The models of CCU-induced liver fibrosis in rats wer used in the experiment.The experimental rats were divided into 6groups: untreated, CCU control, Tetrandrine group, and three HSA groups(containing HSA 1.5g/kg, 0.5g/kg, 0.1g/kg weight respectively). Alanine transaminase(ALT) , aspartate aminotransferase(AST),albumin(Alb) and total protein(TP) in serum and Hydroxyproline(Hyp) in liver were measured. We also observed the changes of liver histopathology to prove the antifibrosis effect of different doses of HSA. (3) Anti-virus experiment invitor: Using the HBV genetic transfection hepatic cell of human-2.2.15 cell as cellular model to screen anti-HBV drugs. To observe the effect of HSA on the 2.2.15 cell secreting HBeAg& HBsAg using SPRIA and test its toxicity on hepatic cell with MTT chrornometry. Anti-acute liver injury: The models of TAA-induced acute liver injury in mice and 5 doses of HSA(2g/kg, 1g/kg, 0.5g/kg, 0.25g/kg, 0.125g/kg)were used to study the protective effect and involved possible mechanisms of HSA on liver injury.
