ObjectiveTo investigate the relationship between refractory epilepsy and the level of pgp and MDR1 mRNA in peripheral blood by flow cytometry and fluorescence quantify PCR,and to discuss the speciality and sensibility of the two means. Methods:In this study,we selected (A)30 patients with refractory epilepsy ;(B)30 patients with epilepsy controlling well;(C)30 healthy subjects.The expression of pgp and the level of MDR1 mRNA were tested by flow cytometry and fluorescence quantify PCR. Results:1 The expression of pgp: The positive rates of pgp in the plasm of group A is 11.9 ± 5.57%, group Bis 0.83 ±0.58%, group C is 0.57 ± 0.57%., The positive rates of pgp of the group A in refractory epilepsy were significantly higher than the group B in epilepsy controlled well(t=10.86,P<0.01); There were no differentiation between group B and group C.2 Detection of MDR1 mRNA by fluorescence quantify PCR:The expression levels of MDR1 mRNA of the three groups is 1.5 × 10~6 ± 2.0× 10~6copies/ml 7.1 × 10~4 ± 9.4 × 10~4 copies/ml and 1.9× 10~4 ± 2.2× 10~4 copies/ml.The level of the group A is significantly higher than the other two groups. 3 Compare of the two test means: The positive rates of pgp in goup A is 93%;A11 the subjects coulde be detected expression of MDR1 mRNA by fluorescence quantify PCR.If enact > 10~5copies/ml as reference value,the positive rate is higher than the rates tested by flow cytometry.
