In this study, nolatrexed hydrochloride polybutylcyanoacrylate nanoparticles (PBCA-NP) were prepared by modern target-oriented drug formulation techniques and emulsion polymerization method. The bio-degradable high molecular weight polymer polybutylcyanoacrylate (PBCA) was used as the carrier, and nolatrexed hydrochloride, a new anti-hepatocarcinoma agent of the thymidylate synthase inhibitor class, was selected as the model drug. By exploring novel ideas, methods, techniques and formulations, results of this study provides valuable scientific and technical imput for the development and advancement of liver-targeted anticancer therapy.The study evaluated several factors that affect the properties of nolatrexed hydrochloride-PBCA-NP, including pH, physical chemical properties of drug, types and amount of surfactants and stabilizers, mixing rate and time, absorption time and temperature. Based on size of particles, entrapment efficiency and drug loading, the factors that are most important were found to be pH, drug concentration, amount of PBCA monomer and stabilizer. The uniform experimental design was applied to optimize the formulation based on these factors.Three batches of sample were prepared using the optimized formulation.
