Objective:We observed the expression of NK cells' receptors NKG2A and NKG2D in the peripheral blood in patients with primary hepatocellular carcinoma and chronic hepatitis B to investigate their relationship and to make a discussion on the clinical value of the detection in NK cells' receptors.Methods:1. Patients and normal controls were recruited: 35 cases of primary hepatocellular carcinoma (PHCC) patients, 33 cases of chronic hepatitis B, 28 cases of normal controls were gathered from health examine center.2. Collecting 2ml blood of each case with EDTA-2K and detecting them within 2 hours.3. 5 marked antibodies were used: anti-CD3-PC5, anti-CD56-FITC, anti-IgG1-PE, anti-NKG2A-PE and anti-NKG2D-PE. 3 groups were involved in this experiment: The antibodies marked by three fluorescence (PC5, FITC, PE) separately were added to each subject, anti-CD3-PC5, anti-CD56-FITC and anti-IgGl-PE were included in the first group; anti-CD3-PC5, anti-CD56-FITC and anti-NKG2A-PE was included in the second group; anti-CD3-PC5, anti-CD56-FITC and anti-NKG2D-PE was included in the third group.4. Detecting them by Flow cytometry (FCM) assay.5. Blood routine of each case was detected by auto blood cell analyzer.6.
