AFM is an advanced apparatus for detection, and it has been widely applied in the cell biology, such as the surface morphology of erythrocyte detecting.In this paper, the erythrocyte membrane has been visualized using AFM in different conditions. So far, there is not related literatures about laser induced cell membrane changes that analyzed by AFM.Some studies illustrate that the erythrocytes which are biconcave shape become thorn-like shape after Ca2+ incubation. However, there are no reports on studying the Ca2+ induced erythrocyte ultrastructural changes using AFM. In this experiment, the erythrocyte incubated with Ca2+, and many holes on the erythrocyte's membrane were observed, which may be Ca2+ channels. Then, our results indicate that erythrocyte membrane thorns which are induced by Ca2+ incubation fade away after laser irradiation (7.529 J/cm2), however, the erythrocyte was not able to return to normal biconcave structure, and our results also indicate that the average roughness (Ra) of erythrocyte membrane increased obviously. The erythrocyte becomes swollen after Ca2+ inc'ubation, and the swollen structures faded away after laser irradiation and resulted in the erythrocyte crimples, ultimately, the Ra of erythrocyte membrane is unavoidablely increased. Together, these results indicate that the damage of erythrocyte membrane resulted from Ca2+ incubation is partly reversible.In chapter 2,3, we treated the erythrocyte with H2O2 or Fenton system in order to simulate the superfluous OH of organism. To simulate the smaller amount of superfluous OH of organism, we treated the erythrocyte with H2O2. As for the larger amount of superfluous ·OH of organism, the Fenton system is used to treat the erythrocyte. In this experiment, to study whether the laser can weaken the ·OH's damage or not, the erythrocyte is incubated with Ca2+ and then irradiated with Laser, the results indicate that ·OH could damage the erythrocyte membrane, and lead to the Ra of membrane increasing, however, the erythrocyte still maintains biconcave shape. Lower intensity He-Ne laser irradiates the damaged erythrocyte, and leads to the Ra decreasing, but Ra can not return to the normal level (1.433 nm). Our experiment...
