Objective: To explore the role of p53 gene deletion and CD44V8-10 abnormal expression in the development of lung cancer, and the application valuate in the molecule pathological diagnosis.Methods: The p53,CD44v8-10 and CD44v10 gene in the lung cancer was detected using Dual-Fish(Fluorescence in situ hybridization) and Competitive RT-PCR (reverse transcription-polymerase chain reaction) technology .we analyze the expression of CD44v8-10 and CD44v10 using Gel-Image Analysis System respectively, Comparing the ratio of CD44v8-10/CD44v10 in malignant group with controls. Results: 1) The deletion rate of p53 gene is 38.28%in lung cancer group and has statistic significance comparing to normal tissue and benign leision (P <0.01). Dection of p53 gene deletion is a diadynamic criteria of lung cancer, with specificity of 87.5 % ,sensitivity of 60.5 % ,and accuracy of 66.41 %; 2) The expression of CD44v8-10 is higher in lung cancer tissue .the ratio of CD44v8-10/CD44v10 in malignant tumor was above 1.00 (3.88 ± 1.87) compared with under 1.00 (0.54±0.20) in benign leision and normal tissue , and the cutoff point of CD44v8-10/CD44vl0 between malignant and benign lesion was 1.00. the ratio has statistic significance between lung cancer group and controls .as a diadynamic criteria of lung cancer with the ratio of CD44v8-10/CD44v10 ,its specificity is 96.12%,its sensitivity 96.85% ,its accuracy 96.48% (P<0.01) ; 3) The combining tests of p53 gene deletion and CD44v8-10...
