Objective:to analysis aberration of chromsome in bladder tumor tissue with FISH, and explore the expression and significance of chromsome between bladder urinary tract epidermal cancer and non- urinary tract epidermal cancer.Method:1.15 normal bladder mucous membrane tissue were colletcted, the aberration of P16 site in the ninth chromsome , the third chromsome, the seventh chromsome and the seventeenth chromsome was analysed with FISH and MCCN was established.2.The expression of the third, the seventh, the seventeenth chromsome and 9p21 in 25 cases for bladder urinary tract epidermal cancer and 13 cases non- urinary tract epidermal cancer(7 cases for squamous carcinoma and 6 cases for adencarcinoma) was dected with FISH, and explore the expression and significance of chromsome between bladder urinary tract epidermal cancer and non- urinary tract epidermal cancer.3.Try to find out applicable qualification and range of FISH's probes of our own country and summarize their advantage and disadvantage and applicable prospect, then establish stanard detective method.Results:1.15 cases of normal bladder mucous membrane tissue were counted, the signal number of the general cell hybridization was analysed. At least 100 nonoverlapping interval cell nucleus were counted in every case, and establish MCCN(general signal number divide cell nucleus number ) to indicate chromosomal multiply. Among 38 samples of bladder tumor detected by FISH's probes, the third, the seventh and the seventeenth chromosome were amplicated obviously. MCCN of chromsome between tumor and normal tissue were respectivly: 3rd 2.43vs1.46, 7th 2.29vs1.44, 17th2.29vs1.30 (p<0.01), gene p16 of 9th 1.36vs1.14 (p=0.05). 2.There was on significant difference in the rate of application including 3rd, 7th and 17th chromosome between bladder urinary tract epidermal cancer and non- urinary tract epidermal cancer. However, the rate of 9p21's deletion had obvius relationship between two of them. The rate of deletion of transitional cell carcinoma, squamous carcinoma and adencarcinoma were respectivly: 52.0%(13/52) 100% 7/7 83.7%5/6 P=0.037 .3.The probes of FISH need more improvement, acidum should be added to handle impurity in the initial stage of cells; the doseage and action time of pepsinogen should be controlled probably; for the denaturation temperature of probe, it can be highered 3-5 centidegrees probably, and take harmfulness of formamide into consideration, therm-platform method was replaced. The results was satisfied.Conclusion:1.FISH is fast, non-trauma and has high detection rate. It can help to explore the relationship between aberration of the 3rd, 7th ,17th chromosome and 9p21 and styles of tumors. It probably also can be useful indicator to diagnosis bladder non-uroepithelium cancer early.2. FISH provides powerful evidence from gene's field to complete differential diagnosis early between bladder urinary tract epidermal cancer and non- urinary tract epidermal cancer and then further clinical treatment is guided better.3.FISH also define that correlated situation of chromosomal abnormality also can be found in bladder non- transitional cell carcinoma through evaluating paraffin wax tissue section including bladder transitional cell carcinoma, bladder non- transitional cell carcinoma and normal urothelium tissue, so it provide probability of proving that detect the genetics change of bladder non-uroepithelium cancer early with sample of urine.4.Further clinical research should be needed to difine whether FISH can be used to complete differential diagnosis of bladder tumors early because of expensive experimental reagent and tedious technology and condition of lab should be controlled at the same time. Stability and reliability of our own country's probes of FISH needed to enhance continually, and it's good to make better fundament to develop clinical medicine. More accurate and integrity conclusion is better for accumulation and analysis of correlated research data in future.
