Effects Of Different Therapeutic Methods And Its Typical Recipes On Activation Of P38MAPK Of Kupffer Cells In Rats With Fatty Liver

Objective: To observe the effects of different TCM therapies: soothing liver, invigorating spleen, dispelling dampness, promoting blood flow and complex recipes on serum lipid, liver function, liver histopathology and P38 MAKP activity of liver Kupffer cell in rats with fatty liver.Methods: The rat model of fatty liver was set by feeding rats with high fat and sucrose diet combined with alcohol. Meanwhile, rats were given recipes of soothing liver, invigorating spleen, dispelling dampness, promoting blood flow and complex recipes respectively. 10 weeks later, we view the influence of different therapeutic ways and corresponding recipes on serum lipid, liver function in rats with fatty liver; observe the histopathological changes of liver under light microscope and electron microscope; explore the impact of different recipes on P38 MAKP activity of liver Kupffer cell. Results:1.Compared with uncontrolled group the contents of serum TC ,TG ,ALT and AST in controlled group significantly increased(P<0.01 or P<0.05).After the intervention of decoction,the contents of serum TC, TG, ALT and AST in each treatment group were lower than those in controlled group and there was a significant difference between soothing liver group and controlled group(P<0.05).2. In controlled group, light microscope observation revealed fatty degeneration from medium to severe degrees in hepatocytes: swelling of hepatocytes and different sizes of lipid droplets inside the cytoplasm. Infiltration of inflammatory cells and much focal proliferation of fibers were also shown in the portal area of controlled rats. Compared with the controlled group, the degrees of fatty degeneration in treatment groups were significantly ameliorated,especially in the invigorating spleen group .Swelling of hepatocytes, infiltration of inflammatory cells and proliferation of fibers in treatment groups were also ameliorated in different degrees.3.In controlled group, electron microscope observation revealed a great deal of lipid droplets, swelling of mitochondria and endoplasmic reticulums in hepatocytes.
