Effects Of Different Therapeutic Methods And Its Typical Recipes On Activation Of ERK1/2 In Kupffer Cells Of Rats With Fatty Liver | | Posted on:2007-12-24 | Degree:Master | Type:Thesis | | Country:China | Candidate:Q Wang | Full Text:PDF | | GTID:2144360212472879 | Subject:Traditional Chinese Medicine | | Abstract/Summary: | PDF Full Text Request | | Aims: To establish the rat model of fatty liver by feeding high fat and sucrose diet combinated with distillate spirit, and to investigate effects of different therapeutic methods and its typical recipes on activation of ERK1/2 in kupffer cells of rats with fatty liver.Methods: The rat model of fatty liver was established by feeding high fat and sucrose diet combinated with distillate spirit.Meanwhile soothing liver, invigorating spleen, dispelling dampness, promoting blood circulation, and complex recipes were given to treat different groups respectively.12 weeks later, the kupffer cells were isolated from livers of normal group, model group and defferent treated groups by sequential in situ perfusion with collagenase and pronase, density gradient centrifugation, selective adherence. The expression of total ERK1/2 and phospho-ERK1/2 in kupffer cells of normal group, model group and defferent treated groups were detected by western blot.Results: In model group, light microscope observation revealed fatty degeneration from medium to severe degrees in hepatocytes. Compared with the model group the degrees of fatty degeneration in treated groups were ameliorated, especially in invigorating spleen group and soothing liver (P<0. 05) . There were significant defference in the inflammatory between normal group and model group. The expression of total ERK1/2 and phospho-ERK1/2 were higher in Kupffer cells from model group compared with normal group. Phosphorylation of ERK1/2 was 9. 8-fold greater in model group compared with normal group. The total ERK1/2 was 6-fold greater in model group compared with normal group. The expression of total ERK1/2 and phospho-ERK1/2 were reduced in all treated groups especially in invigorating spleen group and soothing liver group.Conclusions: This results indicate that SD rats fed high fat and sucrose diet combinated with distillate spirit can develop fatty liver in 12 weeks. Our analyses suggest that the high expression of ERK1/2 and phospho-ERK1/2 involve in fatty liver and contribute to liver injure. The recipes of invigorating spleen and soothing liver maybe protects against liver injury by reducing the total ERK1/2 and activation of ERK1/2 in rats fatty liver.
