Objective (1)To investigate that the destroyed rate of rabbit platelets in liver and lifespan of them after UDP-Gal or trehalose is added into them and to study on the effect of UDP-Gal and trehalose on Platelets Stored at low temperature. To investigate the relationship between dose of UDP-Gal or trehalose and lifespan of platelets. (2) To investigate whether UDP-Gal or trehalose still has protective effect on platelets after longtime storage, that is to say, the relationship between storage time and lifespan of platelets.Methods (1)21 rabbits were divided into seven groups.Each group included three rabbits.10Ml cardiac blood was taken and made into concentrated platelet(CP).CP were stored at four different conditions, such as normal temperature ,low temperature and low temperature+different dose of UDP-Gal for 24h.Platelets were transfused into rabbits'bodies after being labelled with 51Cr and lifespan of 24,48,72h was calculated. In addition, at 72h rabbits were killed and dissected. CPM of each rabbit's liver,spleen ,lung and blood was measured;(2)33 rabbits were divided into eleven groups.Each group included three rabbits.10Ml cardiac blood was taken and made into CP.To add the most suitable dose of UDP-Gal obtained from the test 1 to CP. Then CP were stored for 24 hours,4,6,8,9,10,11,12,13 and 14days.Platelets were transfused into rabbits'bodies after being labelled with 51Cr and lifespan of 24 was calculated. At 72h rabbits were killed and dissected. CPM of each rabbit's liver, spleen ,lung and blood was measured. (3) 10Ml cardiac blood was taken and made into concentrated platelet(CP).CP were stored at different conditions, such as normal temperature ,low temperature and low temperature+different dose of trehalose for 24h.Platelets were transfused into rabbits'bodies after being labelled with 51Cr and lifespan of 24,48,72h was calculated;(2) Each group included three rabbits.10Ml cardiac blood was taken and made into CP.To add the most suitable dose of trehalose obtained from the test 1 to CP. Then CP were stored for 24 hours,2,4,6,8,9,10,11,12,13 and 14days.Platelets were transfused into rabbits'bodies after being labelled with 51Cr and lifespan of 24h,48h and 72was calculated.Results (1)Survival rate of platelets stored at 4℃is lower obviously and radioactivity intake of liver is higher than those of platelets stored at nomal temperature. After different dose of UDP-Gal is added into the platelets stored at 4℃,the destroyed rate in liver decreases and lifespan is prolonged obviously. And with the dose increasing,the lifespan of platelets areprolonged more effectively.But when the dose is more than 5mg·mL-1,the lifespan do not increase and is more than that of platelets stored at normal temperature. (2)UDP-Gal still protect platelet effectively after it has been added into platelets stored at 4℃for 12d is also found. (3)Survival rate of platelets stored at 4℃is lower obviously than those of platelets stored at nomal temperature. After different dose of trehalose is added into the platelets stored at 4℃,lifespan is prolonged obviously. And with the dose increasing,the lifespan of platelets are prolonged more effectively.But when the dose is more than 50mg·mL-1,the lifespan do not increase ,but is lower than that of platelets stored at normal temperature. (4)Trhalose still protect platelet effectively after it has been added into platelets stored at 4℃for 14d.Conclusion Both UDP-Gal and trehalose have a good protection effect on liquid platelets stored at low temperature and still protect platelet effectively after platelets have been stored at 4℃for long time.
