According to Chinese Pharmacopoeia, the drying flower bud of Lonicera japonica Thunb. are used for Flos Lonicerae japonicae in Chinese clinics. It had the functions of heat-clearing and detoxicating, debugging wind-heat and was widely used for the treatment of carbuncle, boss, nail-form boil, sore, throat arthralgia spasm, erysi-pelas, pyretic toxicity and hematodiarrhoea, anemopyretic cold, febrile, and so on.To investigate the active components of"yin qiao herbal pairs", and elucidate the material foundations of pharmacodynamic actions, the chemical constituents of Flos Lonicerae japonicae have been studied by means of modern chromatographic techniques and spectroscopic analysis (1H-NMR, 13C-NMR, HMBC, HSQC, ESI-MS, FAB-MS). 9 Compounds were isolated from the water extract of the flower bud. And their structures were elucidated as epi-vogeloside (shh-1), vogeloside (shh-2), sweroside (shh-3), Loniceroside A (3-O-α-L-arabinopyranosyl hederagenin 28-O-α-L-rhamnopyranosyl-(1→2)-[β-D-xylopyranosyl-(1→6)]-β-D-glucopyranosyl ester, shh-4), 3-O-β-D-glucopyranosyl hederagenin 28-O-β-D-glucopyranos-yl-(1→2)-[β-D-xylopyran-osyl-(1→6)]-β-D-glucopyranosyl ester (shh-5), 3-O-β-D-glucopyr-anosyl-28-O-[α-L-rha-mnopyranosyl-(1→2)-[β-D-xylopyranosyl-(1→6)]-β-D-glucopyrano-syl] oleanolic acid (shh-6), Loniceroside C (3-O-β-D-glucopyranosyl hederagenin 28-O-α-L-rhamnopyran-osyl-(1→2)-[β-D-xylopyranosyl-(1→6)]-β-D-glucopyranosyl ester,shh-7), Loniceroside B (3-O-α-L- rhamnopyranosyl-(1→2)-α-L-arabinopyranosyl hederagenin28-O-α-L-rh-amnopyranosyl-(1→2)-[β-D-xylopyranosyl-(1→6)]-β-D-glucopyranosyl ester, shh-8), chlorogenic acid (shh-9). Compounds 5 and 6 are new compounds.An HPLC method was established for the simultaneous determination of Loniceroside A&C, and 15 crude drug materials were determined. The method was stable, reliable and reproducible. As a result, the contents of Loniceroside A&C are diverse in Flos L. japonica from different regions. Triterpenoid saponin is one of the antibiosis and dephlogisticate compounds in Flos L. japonica. Therefore, the distribution of triterpenoid saponins should be further investigated for Flos L. japonica.
